چکیده

Cytochrome mediates the step in respiratory chain. accepts electrons by one cytochrome c accumulates four equivalents until finally reduces The reducing reside on hemes (cyto(I and u3, resp.) on two There is evidence that electron transfer cytochrome c oxygen crosses cristae membrane mitochondria whereby contributes the electric generation (for review, see [ 1 ] ). The orientation of the two hemes relative to the cristae membrane and the rotational mobility of the whole enzyme is subject to current studies [2,3]. A previous paper [2] reported on the polarized flash photolysis of the complex formed between cytochrome a3 and Co. The linear dichcoism of the resulting absorption changes was analysed to yield information on heme symmetry and rotational mobility of the enzyme. At a given wavelength pair of excitation (585 nm) and of observation (445 nm), a dichroic ratio of about 413 was observed, which was interpreted as indicative of an ‘effective’ 4-fold symmetry of the n-electron system in cytochrome a3. To account for the fact that the same dichroic ratio resulted for the isolated and immobilized enzyme as well as for the enzyme in mitochondria, two alternative interpretations were given: (a) Either cytochrome-c-oxidase is completely immobilized within the cristae membrane, or (b) it carries out only uniaxial Brownian rotation with the rotation axis in parallel to the symmetry axis of the heme (normal to the heme plane).

Toxicity mechanisms of Cigarette Smoke on Eye and Kidney using Isolated Mitochondria

Cigarette smoking is one of the main risk factors for premature human death associated to a variety of respiratory and vascular diseases, and cancer due to containing Hundreds of toxicants. Rat mitochondria were obtained by differential ultracentrifugation and incubated with different concentrations (1, 10 and 100%) of standardized cigarette smoke extract (CSE). Our results showed that cigarett...

Respiratory Enzymes and Mitochondrial Morphology of Hela and L Cells Treated with Chloramphenicol and Ethidium Bromide

Exposure of HeLa and L cells to chloramphenicol causes a progressive dose-dependent decrease in cytochrome oxidase and succinate-cytochrome c reductase activities, concomitant with an increase in the amount of cytochrome c. At 2-3 days, the specific activities of the enzymes have fallen to about one-half of control values; the mitochondria appear swollen. By day 5, enzyme activities are about o...

Cytochrome oxidase in a cytochrome oxidaserich submitochondrial membrane.

1. A cytochrome oxidase-rich submitochondrial membrane (green membrane) was obtained from beef heart mitochondria after extraction of flavoproteins, cytochrome b, Cll C, etc. by treating with deoxycholate and potassium chloride. 2. The green membrane was formed by self assembly from the membrane fragments (flat sheets), which derived from the cristae membrane of mitochondria and had essentially...

Cytochrome c-induced increase of motionally restricted lipid in reconstituted cytochrome c oxidase membranes, revealed by spin-label ESR spectroscopy.

Cytochrome c oxidase isolated from beef heart mitochondria was reconstituted in bilayer membranes of the anionic lipid dimyristoylphosphatidylglycerol (DMPG) with varying enzyme/DMPG ratio. Lipid-protein interactions in the reconstituted membrane complexes were studied in the presence and absence of saturating amounts of bound cytochrome c, by both chemical binding assays and spin-label ESR spe...

Imaging cytochrome C oxidase and FoF1-ATP synthase in mitochondrial cristae of living human cells by FLIM and superresolution microscopy

Cytochrome C oxidase and FoF1-ATP synthase constitute complex IV and V, respectively, of the five membrane-bound enzymes in mitochondria comprising the respiratory chain. These enzymes are located in the inner mitochondrial membrane (IMM), which exhibits large invaginations called cristae. According to recent cryo-tomography, FoF1-ATP synthases are located predominantly at the rim of the crista...
